Characteristical and Identification Study of Bacterial Isolater of Vitamin B12 from Tempeh (Karakterisasi dan Uji Potensi Isolat Bakteri Tempe Pembentuk Vitamin B12)
To promote the quality of tempe (Soybean-bread), especially for its B12 content, a characteristic and identification study of bacterial isolater of vitamin B12 from tempe was evaluated. The procedure used is according to Bergey, the isolate was tested for its potency in liquid medium culture as well as in solid medium culture. Analysis of vitamin B12 was performed according to microbiologycal methods using lactobacillus leichmannii. A total of 82 bacteria isolated various tempe sampler from Java, 8 isolates was characterized and identified as klebsiella pneumoniae and only one isolates as Enterobacter sp bacteria. Result from liquid medium shows that K. pneumoniae isolate can produce vitamin B12, while enterobacter sp is unable to produce vitamine B12. From the 8 isolates producing vitamin B12 only 3 isolates (A15-1, Q7 and LP14) have higher potency, viz producing vitamin B12 more them 3.0 ng/ml. Results from solid medium shows same production pattern on from liquid medium, where isolate A15-1 is highest followed by Q7 and LP17. Inoculation of the 3 isolates in tempe fermentation can increase vitamin B12 content 1.3 ng/g or about 3.25 times of tempe content in general (0.4 ng/g). Futher evaluation is needed for optimal increase of vitamin B12 in tempe and to recognize organic acids preduce by these isolates.
