Pengaruh Perlakuan PGV-1, PGV-0 dan Kurkumin terhadap Protein yang Terlibat dalam Siklus Sel Fase G2-M dan Apoptosis pada Sel Kanker Payudara T47D (The Effect of PGV-1, PGV-0 and Curcumin on Protein Involve in G2-M Phase of Cell Cycle and Apoptosis on T47D Breast Cancer Cell Line)

Previous experiments showed that curcumin analogue (PGV-1) inhibited breast cancer cell (T47D) growth at G2-M phase and induced cell apoptosis. This experiment was conducted to investigate the molecular effect of another curcumin analogue, PGV-0 and curcumin on the cell cycle progression and apoptosis as compared to the PGV-1. Flowcytometric method was conducted to analyze the effect of PGV- 1 (2,5 µM), PGV-0 (5,0 µM) and curcumin (10,0 µM) on the cell distribution ofvarious phase of T47D cell cycle. Western blot was also conducted to observe the effect ofthose compounds on proteins that involved in cell cycle (i.e. p21 and Cdc-2) and apoptosis (Caspase-3/7/9).
The results showed that PGV-1, PGV-0 and curcumin induced hyperploidy phenomenon on T47D cell. PGV-1 inhibited the cell cycle specificallyon G2-M phase. Molecular observation showed that PGV-1 and PGV-0 were able to increase the expressten of p21 protein and the Cdc-2 proteins, whilst curcumin was able to activate the Cdc-2 protein. The compounds have ability to induce apoptosis on T47D cell via Caspase3/7 activation. In conclusion, PGV-1, PGV-0 and curcumin inhibited the T47D cell cycle progression and induced cell apoptosis.

Abstrak
Penggunaan antihipertensi yang tepat mempunyai peranan penting dalam keberhasilan pengobatan hipertensi. Penelitian ini bertujuan untuk mengevaluasi kesesuaian penggunaan obat antihipertensi dan biaya obat pasien hipertensi Umum, Askes dan Jamkesmas rawat inap di IRNA-B RSUP X Jakarta. Penelitian bersifat deskriptif analitis dan dilakukan secara retrospektif dengan kriteria inklusi pasien hipertensi primer dan dengan penyakit penyerta (hypertensive heart disease dengan dan/atau tanpa congestive heart failure, hypertensive renal disease atau renal failure). Hasil evaluasi menunjukkan bahwa ketepatan dosis obat antihipertensi cukup baik yaitu 87,0%. Penggunaan obat di luar formularium cukup tinggi yaitu sebesar 39,3%. Biaya obat antihipertensi per jenis yang paling rendah adalah untuk pasien Askes. Terdapat perbedaan biaya obat antihipertensi antara pasien Askes dengan pasien Umum, maupun antara pasien Askes dengan pasien Jamkesrnas, namun tidak terdapat perbedaan biaya obat antihipertensi yang dikeluarkan antara pasien Umum dengan pasien Jamkesrnas.

Abstract
The appropriate use of antihypertensive have important role for the successful of hypertension therapy. The study aims were to evaluate the use of antihypertensive medicines and cost ofhypertension therapy in General, Askes and Jamkesmas inpatient at IRNA-B ward in X hospital from July to December 20 I 0 period. Design of the study were descriptive analytical and retrospectively. Inpatients with primary hypertension and concomitant disease (hypertensive heart disease with and without congestive heart failure and hypertensive renal disease with renal failure) at IRNA-B ward in X Hospital were included to the study. The results show ed that the appropriateness of the selection of antihypertensive was 87.0%. Non-formulary antihypertensive medicines were 39.3%. The lowest of medicines cost of hypertension therapy was shown in Askes inpatient. There were differences in the cost ofhypertension therapy among Askes with General inpatient and Askes with Jamkesrnas inpatient. There was no diference drug cost therapy between General and Jamkesrnas inpatient.
