Optimasi Formula Floating Tablet Kaptopril Sistem Effervescent Menggunakan Desain Faktorial (Optimization Formula of Captopril Floating Tablet Efferveascent System Using Factorial Design)
Captopril has only 6-8 h duration of antihypertensive action, most stable at acidic condition and as the pH increased, it becomes unstable and undergoes a degradation reaction.These facts indicate a promising potential of the captopril floating system as an alternative to the conventional dosage form.The objective of the recent investigation was to find an optimum formula of floating tablet for captoprH using direct compression technology. A 2' factorial design was applied to systematically optimize the formula. The amounts of sodium bicarbonate (XA), citric acid anhydrous (XB), and HPMC K4M (Xc) were selected as independent variables. Floating lag time and dissolution efficiency (DE'Bo) were selected as dependent variables. The floating behaviour and in vitro dissolution studies were carried out in a USP paddle apparatus in buffer solution at pH 1.2 at 37°C. Based on contour plot of both responses, the optimum formula contains sodium bicarbonate 40-55 mg, citric acid 20-40 mg, and HPMC K4M 120 mg.
