The Effect of Cellulose Derivatives on Lynestrenol Release Rate from Nicroencapsulated Dosage from in Vitro-in Vivo Correlation (Pengaruh Derivat Selulose terhadap Kecepatan Pelepasan Linestrenol yang Dimikroenkapsulasi dan Korelasinya secara In Vitro-In Vivo)
The aim of this research is to obtaine a sustained release dosage form with cellulose derivatives by microencapsulation method. In this study, the microencapsulation method modified from Lordi, by spraying the ethanolic solution of methylcellulose, ethylcellulose and hydroxyprapyl methylcellulose. Particles of the active substance were put in the coating pan and sprayed carefully with the solution of those celluloses that had been swollen in ethanol 95 percent. The granulules were dired and were then shieved on 30/40 mesh. The dissolution test was carried out in the medium of phosphate buffer at pH 1.2 and pH 7.6. Another portion of the granule was tested in vivo for the absorption rate on fasting rabbits. The concentration of all samples were determined by spectropotometric method. Microencapsulation dosage from by ethylcellulose exhibited the slowest of active substance release followed gradually by hydroxypropyl methylcellulose and methycellulose. This suggest that the length of side chain of the cellulose derivatives might effected drug release rate in vitro as well as in vivo.
