Study on biological activities of endotoxin and lipid A, and structure elucidation of lipid A has been conducted by researchers in many countries, especially in Japan, Germany, and USA. The synthetic approaches and the biological activity test of lipid A showed that the proposed-synthetic lipid A had the same activities with the natural lipid A from . It has also been recognized that lipid A seemed to be the active centre and responsible to endotoxin activities of gram negative bacteria. Furthermore, the biological activities test on many synthetic lipid A analogs containing different number and combination of , showed that fatty acid moieties have an important role in the endotoxin activities of gram negative bacteria. Studies on immunogenicity and antigenicity of lipid A or LPS can be done using synthetics lipid A or LPS-labelled with 14C.
