The Effects of High Level of Fats and its Source in the Diets on the Toxic or Carcinogenic of N-nitrosodimethylamine and Ethynyl Estradion on the Lung of Experimental Rat (Pengaruh Diet Berkadar Lemak Tinggi terhadap Efek Toksin atau Karsinogenik Inisiator N-nitrosodimentilamin dan Promotor Entilin Estradiol pada Paru Tikus Wistar)
This experiment was designed to study the effects of high level of fats and its source in the diets on the toxic or carcinogenic effects of N-nitrosodimethylamine (NDMA) and ethynyl estradion (EE) on the lung of experimental rats. This experiment was done in 72 rats given NDMA and or EE and fed diets of normal fat level or 15 percent fats, consisted of 10 percent animal or vegetable fats. Rats were killed at 135 days after promotion, then lungs were put into 10 percent formalin. Tissue samples were stained with the method of hemmatoxylin and eosin (H
E). Macroscopic examination revealed the presence of lung nodules in group of rats given NDM, NDMA and EE or EE. Addition of 10 percent animal or vegetable fats in the diets were found to have a greater effects in increasing the number of lung nodules compared to groups fed normal diets. The source of fats were not have a different effects on the development of lung nodules. Microscopic examination revealed that the lung nodules were classified as adenoma which might be orginated from alveolar epithelial cells. Results indicate that diets with additional 10 percent animal or vegetable fats showed a greater effects on the lung carcinogenesis compared to normal diets.
