Therapy of Asthma Using Slow Released Theophylline in One Day Singel Dose (Pengobatan Asma dengan Teofilin Lepas Lambat Dosis Sekali Sehari)
The efficacy abd tolerability of (Pirasmin-Retard) was compared with theophylline 4x150 mg in a cross-over study involving 30 patients with asthama bronchiale. Theophylline were essayed on day-7 (after 7-days administration of Theophylline 4x150 mg); on day 14 (after 7-days administration of Pirasmin-Retard 650 o.d) and on day-21 (after 14-day administration of Pirasmin-Retard). The means were day-7 : day-14 = 2.6 plus/minus 6.2 mcg : 7.0 plus/minus 7.7 mcg (p=0.001), and day-7 : day-21 = 2.9 plus/minus 5.7 mcg : 7.3 plus/minus 6.9 mcg (p less then 0.001). Pulmonary function test (FEV-1, FVC, PFR) were performed and the clinical symptoms (cough, disturbances of activities and sleeps, wheezing) were also evaluated. The result confirmed that the administration of Pirasmin-Retard (once-a-day Theophylline) provided significant better theophylline plasma levels. The measurement (FVC and FEV) increased very significantly but there were no significant difference between group A and B.
