Efek Catechins Teh Hijau terhadap Ekspresi ROR α dan C/EBP-α pada Kultur Primer Preadiposit (Effect of Green Tea Catechins on ROR α and C/EBP-α Expression on Preadipocyte Primary Culture)
Obesity is o major risk factor for most of chronic diseases with high mortality. Catechins, the active constituent of green tea has been regarded beneficial as an anti-obesity. This study was aimed to investigate the effect of catechins on the positive and negative regulators of lipo genesis and odipogenesis; ROR-a and C/EBP-α.
This study was performed in primary cultured of preodipocyte divided into 4 treatment groups: (1)control, (2) catechins 5 µM, (3) catechins 10 µM and (4) with catechins 30 µM. Two days post confluence, cell were induce to differentiate into adipocyte with IBMX, DEX and insulin. On days 3 of differentiation, cells were treated with catechins for 24 hour Cells then harvested and measurement of ROR-α and C/EBP-α were performed using ELISA and immunocytochemistry.
Result show that catechins significantly reduced the expression of C/EBP-a by 20,24% don 26,43%. it was identified that found that ROR-a expression increased, but not reach statistical significance. These results suggested that cotechins at leostin part inhibit adipogenesis through C/E8P-α.
