Pengaruh Propylthiuouracil terhadap Kadar Testosteron dan Estrogen pada Tikus Sprague Dawley Betina
Iodine Deficiency Disorders (IDD) was associated with female reproductive hormones disorders. Hypothyroidism inhibits follicular development which subsequently inhibited estrogen secretion. Biosynthesis of estrogen derived from testosterone through aromatization mechanism in ovarian granulose cells.
This study aimed to determine changes in levels of serum testosterone and estrogen in hypothyroid rats induced by propylthiouracil. This study was an experimental post test only with controlled group design
Study used 11 Sprague dawley female rats aged 2.5 months with an average body weight 111.1 grams. Five rats given propylthiouracil in the drinking water with doses of 0.1 g/L for 30 days and six rats were used as controls. Hypothyroid rats were determined by measurements of free T4 levels. Data of free T4 levels, testosterone, and estrogen taken from blood serum and it were measured using ELISA technique. Data of testosterone, estrogen, and free T4 analysis using independent sample t test. Free T4 levels of treatment group were 4.0 ± 0.4 pmol/L and the controlled group were 8.1 ± 1.6 pmol/L, statistical analysis showed significant difference between the two groups with p values = 0.001. Testosterone levels of the treatment group were 0.72 ± 0.1 pg/dL and control group were 0.66 ± 0.1 pg/dL, statistical analysis showed no significant difference between the two groups with p values = 0.20. Estrogen levels of the treatment group were 9.4 ± 6.2 pg/dL and control group were 12.6 ± 3.9 pg/dL, statistical analysis showed no significant difference between the two groups with p values = 0.35.
There was no significant change in the levels of testosterone and estrogen serum in Sprague dawley female hypothyroid rats induced by propylthiouracil.
