Optimasi Teknik Isolasi DNA dan RAPD - Pf.R pada Keong Oncomelania hupensis lindoensis sebagai Hospes Perantara Schistosomiasis di Sulawesi Tengah (Optimization Techniques and lsolation DNA RAPD - PCR Oncomelania Hupensis Lindoensis Snail As Intermediate Host of Schistosomtasis in Central Sulawesi)
ln lndonesia schistosomiasis is caused by Schistasoma japonicum with O.h.lindoensis snails as an intermediate host. For that we need to do research on the level of DNA polymorphism Oh.lindoensis snaiis. One method used is the technique of Random Amplijied Polymerase Chain Reaction (RAPD-PCR). Therefore we need a proper method of DNA extraction as well asin determining the optimum reaction conditionsfor RAPD-PCR. This study aimed to be obtained of Oh.lindoensis DNA from in the high lands of Central Sulawesi in Bada, Napu and Lindu and to optimize condition ofPCR-RAPD. To be obtained Oh.lindoensis DNA using with protocols or operating procedures PureLink Genomic DNA Mini Kits TM. The extraction ofDNAyieled goad quality ofDNA While ampliflcotion of DNA using PCR-RAPD will become efficient and consistent if the amplification reoctions are in ideal condition. In O.h.lindoensis, clear PCR-RAPD patterns were obtained using lOng DNA template, 25 pm 01 primer and the number of thermal cycle was 40 x.
