Pengaruh Komposisi Media terhadap Aktivitas dan Karakter Enzim Tanin Asil Hidrolase dari Aspergillus niger (Effect of Media Composition on Activity and Character of Tannin Acyl Hydrolase from Aspergillus niger)/
Abstract: This research aimed to investigate the effect of medium compositions in production of tannin acyl hydrolase (tannase) from Aspergillus niger. The parameter investigated were ratio of wheat ftour and Czapeck medium, and volume of inoculum. Ratio of wheat ftour and Czapeck medium were 1: 1; 1:2; 1:3; 1:4 and 1:5 (w/v) and volume of inoculum were 1, 2 and 3 mL. The tannase product was fractionated and characterized. The result showed that optimal tannase product was obtained at ratio of wheat ftour and Czapeck medium 1:2 (w/v) with inoculum volume of 1 mL. The best specific activity of tannase was obtained at saturation level of 70%. Characterization results showed that optimum activity was at 40°C and pH 6. Tannase was increased by the presence of K+and Na+. The presence of metal ions such as Zn2+, Cu2, Ca2+, Mn2+, Fe2+ and Mg2+ was known to inhibit the enzyme activity.
