PENGARUH PEMBERIANMINYAK JINTAN HITAM (Nigella sativa)TERHADAP KADAR ALT DAN AST PLASMA TIKUS PUTIH (Rattusnovergicus)MODEL HEPATOTOKSIK (ETANOL)
The active constituents of the volatile black seed oil (Nigella sativa),ymoquinone, was found to be hepatoprotective against ethanol induced hepatotoxicity via its antioxidant mechanism and anti-inflammatory features. This research was aimed to reveal whether ethanol influence on the plasma ALT and AST level, whether black seed oil influence on the plasma ALT and AST level of hepatotoxic (ethanol) rats
model, and also, how much black seed oil effective dosage that can decrease the increasing plasma ALT and AST level of hepatotoxic (ethanol) rats model. With experimental method and completely randomized design,24 male wistar rats (Rattus novergicus) were divided into 4 groups. Group I only received a single dose of
ethanol 2 ml/100gr body weight orally, while group II, III and IV received 0,5, 1, 2 ml/Kg body weight/day of black seed oil for 7 days then ethanol 2 ml/100gr body weight orally was induced. Plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured before and after black seed oil and ethanol administration. Data analysis methods were conducted with paired T test and one way
ANOVA which was followed by post hoc Tukey’s HSD. The result reveals that ethanol caused significant increase in plasma ALT level (p=0,000) and AST level (p=0,000). The use of black seed oil in various dosage (0,5, 1, 2 ml/Kg body weight) caused difference signi ficant increase in plasma ALT level (p=0,000) and AST level (p=0,000) of hepatotoxic (ethanol) rats model. 0,5, 1, 2 ml/Kg body weight of black seed oil was not the effective dosage that can decrease the increasing plasma ALT and AST level of hepatotoxic (ethanol) rats model
