Uji Aktivitas dan Mekanisme Penghambatan Minyak Atsiri Daun Sirih (Piper betle, Linn) dan Ekstrak Etanol Daun Sirin terhadap Beberapa Bakteri Gram (+) (Anbacterial Test of Several Gram (+) Bacterial and the Mechanism of Inhibition in Essential Oil and Ethanol Extract of Betle Leaves (Piper betia, Linn)
Piper betle L is a well known plant used in taditional medicine, espesially used as an antibactteria. The aim ot this research is to compare antibacterial activity and mechanism of inhibition between esential oil etanol 70% extract of betle leaves on some gram (+) becteria. The experimental result showed that MC values for essential oil against Staphylococcus aureus is 7,4% (b/v), Bacillus pumilus is 9,3% (b/v) and bacillus careus is 10,2% (b/v), to Streptococcus viridans and Bacillus subtilis is more than 13,9% (b/v) respectively. The MIC values of ethanol 70% extract of betel leaves against Staphylococcus aureus is 15% (b/v). Bacillus pumilus is 8% (b/v) and Bacillus cereus is 10% (b/v), to Streptococcus viridans and Bacillus subtilis more than 12% (b/v) Mechanism of inhibition of the essential oil and ethanol 70% extract of betel leaves can cause damage and leakage of wall and membrane of tested bacterail cells. These phenomena ware indicated by elevating the Ca and K ion content in medium which was analyzed by atomic absorption spectrophotometry (AAS). In the exeriments, elevating the protein and nucleic acid content in medium which were analyzed by ultraviolet spechtrometry (UV) is also detected in the medium. Finaly, the SEM photograph of treated cells is cleanrly support the cell surface damage and leakage on the cell surface.
