The effective contracception for men should meet a number of requirements: safe, reversible, easy used, quickly wark, good for user and it doesn’t bring had impact for the sexual function. The alternative medicine/therapy for contraception which is good for men is not only limited on hormonal ones. However, it can be obtained from certain plants, which may contain anti-fertility substances. In order to study about the medicine taken from plants, a research has been done on piper betle linn’s extract. This study aim was to find out the influence of betel extract on the spermatogenesis to the disorder of seminal tuble. Alkaloid contained in the extract piper betle linn can inhibit aromatize enzymes that catalysis the conversion of androgen to estrogen which in then increases testosterone. The high concentration of testoesterone has negative feedback to hypophysis, that is holding release of FSH or LH and tannin is able to bind with proteins and reduce its size, which in turn inhibits the spermatogenesis. This was a randomissed pretest posttest control group design consisting of 3 group 1 was the tween 1%. Group II the musculus treated with 500 mg/kg BB/per day and Group III mus musculus treated with 1000 mg/kg BB/per day. After 35 days, the mice were killed: the testis organ was taken, then checked microscopelly. This is aim at knowing the concentition of the sperm and the decay destruction of seminal tuble. The research found that there was a significant decrease of all stadium of spermatogenesis, that is spermatogensis A (p=0.008). spermatosit primer pakhiten (p=0.000) and spermatid (p=0.00). Also, there is significant destruction on seminal tubule, third and fourth category. The third category is intermediate cell and the fourth is the decrease of spermatogenesis
