Perbandingan Kadar Regulatory T-cells antara Kehamilan Normal dan Abortus
To find the role of Treg in maintaining sucessful pregnancies by measuring Treg level in normal pregnancy below 20 weeks, measuring Treg level in miscarriage and comparing the Treg level between normal pregnancy dan miscarriages. An observational, non-intervention, comparative cross=sectional study, was observational, non-intervention, comparative cross-sectional study, was conducted at pergnant female with normal pregnancies and miscrarrriages who come to place of study. Data are collected since the subject come to place of study as long as the subject is within the inclusive criteria and has agreed to sign up the informed consent to join the study. Blood sample is taken intravenously from subject for 50 micro-liters and put through the flow-cytometry with specific reagent for CD45 (PerCP-347464), CD4 (SIPC-340133) and CD25 (PE-34009) and Treg counts are counted with lyse no washed system and Cellquest Pro software. Output from the flowcytometry are then saved and noted. Subjects with normal pregnancies are followed up until 20 weeks of their pregnancies to escure there are no miscarriages up to than time. The subjects with normal pregnancy during the test period then have miscarriages afterwards, before 20 weeks, will then put into the miscarriage group of subjects. From the mean comparison statistic calculation in CD45 cell precentage, it is found that there are no statistically-significant difference between the two groups, where the mean for case group is 10.96 ± 6.57% cell compared to the means of 9.60 ± 5.30% cell in the control group, with p = 0.6.10. In the comparison of Treg level, the statistic calculation found that the median of case group (2.45 sel/µl) compare to median of control group (2.53 sel/µl) didnot show statistically significant difference with p=0.946 (Mann Whitney correlation test). This study found out that there are no significant relations between Regulatory T-cells (Treg) level with miscarriage.
