The Influence of Lysozyme to the Antibiotic Activity of Sephalosphorin Group (7-ACA) and Betha-Lactam (6-APA) [Pengaruh Enzim Lisozim terhadap Aktivitas Antibiotika Golongan Sefalosporin (7-ACA)]

Lysozyme has an ability to degrade some substrates of carbohydrate derivative, lipid and protein, it might also capable to degrade membrane cell of bacteria as well as antibiotics. Therefore the influence of lysozyme toward antibiotic activity had been investigated. The experiments performed using 7-Amino Cephalosporanic Acid (7-ACA) group, (sephalexin and sephradin) and 6-Amino Penicillanic Acid group or 6-APA (Penicillin G and V). The bacteria tested were Gram positive (Staphylococcus aureas) and Gram negative (Escherichia coli). Firstly experiment was done by combining antibiotic with enzyme incubated for 20 minutes and then adding with bacteria and incubation was performed until 18 hours, this treatment is called combination I. Secondly the bacteria (Gram negative and Gram positive) was mixed with enzyme, and incbated for 20 minutes, prior the addition of antibiotic, and incubation was continued for 18 hours, this treatment is called Combination II. Other experiment, was also done by treating bacteria directly with the antibiotic and incubated for 18 hours. The result of observation is presented as MIC (Minimum Inhibition Concentration) and MBC (Minium Bactericide Concentration). The result indicated that lyzozyme addition increased antibiotic activity. Lysozyme up to 400 ug/ml did not show the MIC. Lysozyme is not degrade antibiotic because MIC and MBC at Combination II did not significantly differ from combination I. The MIC and MBC value of Combination II is lower then those of pure antibiotic, indicated that enzyme could only lysis of membrane cell bacteria but did not degrade antibiotic.
