Effect of Supernatan Produced by Mouse Spleen Culture toward Splenocytes, Thymocytes and Bone Marrow Cells (Pengaruh Supernatan dari Kultur Sel Limpa Mencit terhadap Sel Limpa, Sel Thymus dan Sel Sumsum Tulang)
The purpose of this study is to know the effect of growth factors on target cells, such as splenocytes, thymocytes and bone marrow cells. The growth factors were obtained by induction of spleen lymphocyte cells with concanavalin A. These factors were then put into a 96-well flat bottom plate containing different target cells. After 72 hours incubation, H-Tdr were added and the plate was placed at 4 degree C for 6 hours. The cells were harvested on a Whatman paper and dried overnight at room temperature. The activity of the cells assessed for their thymidine radioactive incorporation by using Beta-Emission Counter. The results showed that there were an increasing mitosis index of the cells which received 20 ul growth factor. In conclusion, the growth factors within spleen cells culture supernatan enhanced the mitosis index of the target cells: and the supernation possibly contained II,-2. II,-4 or II,-5 and GM-CSF or II,-3.
