Neutral Gen Protease Isolation from Bacillus megaterium Using Polymerase Chain Reaction Tehnique (Isolasi Gen Protease Netral dari Bacillus megaterium Menggunakan Teknik Polymerase Chain Reaction)

The isolation and characterization of chromosomal genes and bacterial plasmids are important for strain improvement to produce most valuable compounds on industrial scale. The isolation of the neutral protease (npr) fragment from Bacillus megaterium was carried out by polymerase chain reaction (PCR). The PCR experiments were done setting up of variation for oligonucleotide primers, annealing temperature, primer and template concentrations, and number of thermal cycling. Based on the variation of condition for PCR reaction, we found three npr-fragment, 60 bp npr-fragment, 650 bp npr-fragment, and 760 bp npr-fragment.
