Pengaruh Ekstrak Sambiloto (Andrographis paniculata Ness) terhadap Kadar Histamin Srum dan Gambaran Histologis Saluran Pernapasan Mencit Balb/C Model Asma Alergi (The Effect of Sambiloto (Andrographis paniculata Nees) Leaves Extract to the Level of Histamine Serum and Histological pattern of Respiratory Tract in Balb/C Mice Model of Allergenic Asthmatic)
It was reported in previous studies that leaves extract of Andrographis paniculalata Nees possessed pharmacological activities such as anti-inflammatory and histamine inhibitor. It is also known that histamine serum level and respiratory tract inflammation are increasing in asthma condition. The aim of this study was examine the effect of A. paniculata leaves extract to the level of serum histamine and the histological structure of bronchial laver in mice model of asthma. Male Balb/C mice were sensitized and challenged intraperioneally (p) with ovalbumin (OVA). Mice were immunized 1.p. on days 4 and 20with 2.5 mg of OVA adsorbed to 7.75 ml of alluminium hydroxide gel. OVA challenges (10 mg in 1 ml of PBS) were administered aerosolly on days 26, 30, 33, 39 and 46, with mice being sacrificed on day 47. Hatamine level in serum was determined by using ELISA kits. Bronchial microscopic slides were prepared using paraffin method and stained with haematoxyllin easin staining and histopathlogical scoring was based on a scale described by Myou et al, that ranging from 0 to 4. The data were analyzed by ANOVA followed by LSD Test histamine serum levels, whilst for the histological structure using Kruskal-Wallis, followed by Mann-Whitney test. The results showed that A. paniculata leaves extract could decrease the histamine serum levels and minimized inflammation of bronchial layer in the experimental animal.
