Isolasi Senyawa dan Ujiantibakteri Ekstrak Etilasetat Rumput Mutiara (Hedyotis carymbosa L (Lamk) terhadap Bakteri Escherichia coli (Isolation and Antibacterial Test compound of Pear Gras (Hedyotis corymbosa L (Lamk) Ethyl Acetate Extract on Escherichia coli Bacteria)
An experient was conducted to determine antibacterial compound from pearl grass (hedyotis corymbosa L (Lamk) on Escherichia coli bacteria. The experiment was carried out by isolating alcoholic antibacterial compound from ethyl acetate extract of Pearl grass (root, stem, leaf, flower and fruit). Methods in this experiment were laboratory experiment included extraction, isolation, characteristic of the structure and antibacterial activity test. Fractionation and purification of the isolated compound were conducted by chromatography method. Characteristic (elucidation) structure of the isolated compound were analyzed based on prectroscopic data IR ¹H and ¹³C NMR. Antibacterial activity test was carried out by Disk Diffusion Method. The result of IR spectra showed the presence of hydroxyl group (v3340, 5 c -¹), of aliphatic C-H (v 2846, 7 cm-¹), C=C (v 1608,5 cm-¹) and C-OH (v 1060,8 cm-¹). Spectra of ¹H-NMR showed that there were 1 signal for alylic proton at chemical shift (§H) : 3,63 ppm (t), 2 signal for vinylic proton at chemical shift 5,16 (d) and 5,24 ppm (dd). Spectra of ¹³C-NMR showed that there were 6 kind of carbon 2 signals for vinylic carbon were showed §c 122,4 and 128,5 ppm. Based on the data it is suggested that the compound is 4-metil-2en-heksanaol. This compound showed significant antibacterial activity against Escherichia coli.
