Pegaruh Waktu Penyimpanan terhadap Perubahan Kadar Kreatinin, Kalsium Fosfat Urine
To examine uantitative urine and creatinine clearance, 24-hour urine was used as sample, Storage in Room temperature without presentative may result in urine decomposition, leading to the less accurate result of examination. This study was udertaken to discover effect of storage method on the change of creatinine, calcium and phosphate levels in urine stored for 24 hours. The types of treatment used in this study were storage at room temperature, room temperature + HCI, temperature of 2-8 drajat celsius and 2-8 drajat celsius + HCL. Fresh urine sample were obtaine from 30 patient who met the designated criteria. Sample were taken using selective sampling method. As a using atest strip. The results were analyzed by kruskal Wallis non-parametric test, and followed by mann-Whitney Test. Effect of strorage method on the change of creatinine level resulted in p=0,011, indicating that there was a singnificant difference among 4 methods of storage. Storage at room temperature was significanttly diferent from storage at room temperature + HCL (p=0,047), and significantly different from storage at the temperature 0f 2-8 °C (p=0,002) and at 2-8°C + HCI (p=0,015). Efect of storage method on the change of urine calcium level produced a p=0,015,indicating a significant diference among the 4 methods of storage. Storage at room temperature was significantly different from storage at room temperature + HCl (p=0,005), at 2-8°C (p=0.017) and 2-8°C + (HCl (p=0,011). Effect of storage, methods on the change of urine phosphate level also showed significant difference (p=0,08) among the 4 methods of storage, and significantly different from storage at 2-8°C + HCl (p=0,011). Effect of storage at room temperature + HCl (p=0,005), at 2-8°C (p=0,017) and 2-8°C + HCl (p=0,011). Efect of storage methods on the change of urine phosphate level also showed significant diference (p=0,018) among the 4 methods of storage, and significantly diferent from storage at 2-8°C (p=0,37) at room temperature + HCl 9p=0,023), and at 2-8°C + HCl (p=0,003). The average change of creatinine, calcium, and phosphate levels showed that storage at temperature produced higher average level of reduction. To assure the uality of laboratory examination results, it is recommended not to store urine at room temperature for 24-hour without preservative.
