Aktivitas Antiflamasi Kuersetin-3-Monoasetat, Hasil Asetilasi Selektif Kuersetin
Quercetin is a flavonoid having anti-inflammatory activity by cyclooxygenase -2 inhibition. In addition quercetin has a cytoprotective activity against gastric ulcer due to its antioxidative activity. The objevtive of this research was to obtain quercetin derivative through selective acetylation and to find out the anti-inflammatory activity and gastric ulcer side effect of the synthesis product. Quercetin was acetylated with acetic anhydride in dimethylformamide with sodium acetate as catalyst at-10ºC. The product was purified by preparative thin layer chromatography and characterized by melting point measurement, thin layer chromatography, ultraviolet and infrared spectroscopy as well as H-NMR spectroscopy. Anti-inflammatory activity of the product was tested on carrageenan induced rat paw oedema. The result of characterization indicated that the mayor product was quercetin-3-monoacetate. At equivalent dose of 10 mg.kg bw quercetin, this compound showed higher anti-inflammatory activity com-pared with its lead compound, with gastric ulcer less severe than sodium diclofenac of 5 mg/kg bw dose. This result confirmed that monocacetylation could enhance the anti-inflammatory activity of quercetin.
