Chlorofor Fraction of Areca Catechul) Induces Apoptosis and Decreases BCL-2 Expression on MCF-7 Cells
Areca (Areca catechu L.) seeds ethanolic extract shows cytotoxic activity against MCF-7 cells. The aim of this research is to examine the cytotoxic effect, apoptosis induction andtarget on bcl-2 expression in MCF-7 cells of A. catechu chloroform fraction (ACF). The A. catechu seed powders were extracted by using ethanol 96%. Then, the extract was fractioned with partition by hexane, chloroform, and ethyl acetate to collect hexane (AHF), choloform (ACF), ethyl acetate (AEF), and water fractions. Cytotoxic activity was examined by MTT assay. Apoptosis examination was carried out by using acrydine orange-ethidium bromide staining (double staining). The BCI-2 expression was examined using imnunocytochemistry assay. Single application of AHF, ACF and AEF inhibited cell growth on MCF-7 with IC50 of 78 µg/mL 65 µg/mL, and 98 µg/mL (P
