Antibodi Pregnancy Specific Protein B (PSPB) sebagai Bahan Bioaktif untuk Diagnosis Kebuntingan Dini Sapi Perah Frieshian Holstein *FH) dengan Metode Mikrotiter Strip (Antibody Pregnancy Specific Protein B (PSPB) as Bioactive Substance in Early Prgenancy Diagnosis of Frieshian Holstein (FH) Dairy Cattle with PSPB)
The development of early diagnosis method in livestock can be undertaken by observing specific substance presents in the blood of pregnant livestock, i.e. the Pregnancy Specific Protein B(PSPB). The objective of this was to prove that PSPB protein was able to induce humoral immune response characterized by the formation of anti PSPB protein was able that anti PSPB produces was able to recognized PSPB protein for pregnant Frieshian Holstein (FH) dairy cattle blood as the based for developing one of early test in FH dairy cattle. This study was conducted by following these stages: 1) Characerization anti PSPB produces from the induction of PSPB protein isolate from the blood serum of pregnant FH dairy cattle, 2) Pregnancy diagnosis of FH dairy cattle with PSPB microtitre strip. Anti PSPB titre obtained in this study was found to be low in week 5 after the first and second reinjection (week 10), i.e., 47.14 µg/ml and 71.43µg/ml, while the highst level was obtained in week 3 after the first and second reinjection (week 8), i.e., 562.15 µg/ml and 610.00µg/ml. The result of PSPB microtitre strip with abdominal palpation revealed sensitivity, specificity, accuracy, positive predictive value, and negative predictive value of 93.75%, 94.44%, 94.00%,96.77% and 89.47%. Based on the results, it can be concluded that PSPB protein humoral immune response by the formation of anti PSPB produced can be employed for FH dairy cattle pregnancy diagnosis PSPB microtitre strip.
