Background: Piper retrofactum Vahl has been used as traditional medicine for a long time. Objective: to compare: (1) piperine alkaloid and other alkaloid of cabe Jawa from Madura and Lamongan, by Thin Layer Chromatography (TLC) method,(2) the effective dose (ED50) of anticonvulsive of infusum extract and mefenemic acid by shock seizure (MES) and metrazole seizure (MET) in Mice, (3) ED50 of analgetic effect of infusum extract and mefenemic acid by electric stimultion, and prostaglandin (PG) synthesis by measuring malonaldehid (MDA) production in guinea pig pulpae. Results: (1) Piperin alkaloid of cabe jawa from Madura and Lamongan are 3.23 percent, both extract have similiar alkaloid content, (2) ED50 anticonvulsive of extract by MES and Met method is 47.40 (42.27-63.87) and 28.40 (15.30-71.09) mg/kg BW peroral; diazepam is 9.72 (5.0-12.0) dan 0.10 (0.01-0.67) mg/kg BW per oral, (3) ED50 analgesic effect of extrat and mefenemic acid is 15 gram/kg BW and 16 mg/kg BW pe oral, (4) the production of MDA after taking extract is 0.569 ug/20 mg (not significat difference, 't' test, p more than 0.05, after mefenemic acid is 0,088 plus/minus 0.023 ug/20 mg (significant difference, p.0.05). Conclusion : (1) piperine alkaloid content of cabe Jawa from Madura is higher than Lamongan, (2) anticonvulsive potency extract is lower than diazepam, but have similar profile, (3) the extract have a weak analgesic effect and sintesa PG inhibition.
