The Effect of Cimetidine Co-administration on the Renal Elimination of Sulfadiazine (Perubahan Klirens Ginjal Sulfadiazin karena Praperlakuan Simetidin)
The effect of cimetidine co-administration on the renal elimination of sulfadiazine, has been investigated in eight healthy male volunteers. Single dose of 400 mg cimetidine coadministration, increases significantly the renal clearance of sulfadiazine from 12.01 plus/minus SEM 1.82 ml/minutes to 16.41 plus/minus SEM 1.93 ml/minutes (p less than 0.05). The increase in sulfadiazine clearance correlates significantly with intial renal clearance (r=0.7708, p less than 0.001). Suprisingly, cimetidine also increase the acetylation ratio of sulfadiazine from 0.22 plus/minus SEM 0.02 to 0.31 plus/minus SEM 0.03 (p less than 0.05). Therefore this study demonstrates that cimetidine increases both the renal elimination and hepatic acetylation of sulfadiazine.
